Abstract
Approximately 285 million people worldwide are affected by retinal diseases, causing vision loss or even blindness. The loss of photoreceptors is one of the main causes for those that lead to blindness, such as age-related macular degeneration and retinitis pigmentosa. Currently, there is no effective treatment for those retinal degenerative diseases.
Among all different attemps, optogenetic has shown a great potential for functional visual restoration. By introducing light-gated ion channels to the remaining cells in the retina, it not only allows high spatiotemporal resolution but also the possibility to target specific cell types.
Here I present the photostimulation of rod bipolar cells expressing channelrhodopsin-2 in a photoreceptor degeneration mouse (rd10) model while simultaneously recording the retinal output using a CMOS-based microelectrode array. The intrinsic property of rod bipolar cells allows the optogenetic stimulation for both ON and OFF pathways. The distinct ON and OFF activation patterns and the intriguing temporal and spatial resolution give important insights for understanding the performance of vision restoration using optogenetic actuation.
